New germacranolides from Isocarpha species

The investigation of two Isocarpha species has yielded eight new germacranolides most of them belonging to the heliangolides. In addition to known p-hydroxyacetophenone-derivatives, a new dihydroeuparine derivative was isolated. The chemotaxonomical aspects are discussed.     

Nitrate assimilation in Crotalaria juncea (Linn.) pollen suspension culture

In vivo and in vitro activities of nitrate reductase were assayedin Crotalaria juncea pollen suspension cultures. This enzymewas found to be substrate-inducible and enhanced activity wasobserved when it was extracted with cysteine buffer or incubatedwith NADH (0.6 mM) at 25?C or when the germinated pollen grainswere treated with red light for 10 min. Enzymes of ammonia assimilation,glutamate dehydrogenase and glutamate synthetase, and also thepentose phosphate-shunt enzyme, glucose-6-phosphate dehydrogenase,which catalyzes the step that provides reducing power to thesystem, are described. Effect of 7-azatryptophan on heterocyst differentiation inAnabaena doliolum Bharadwaja

The effect of DL-7-azatryptophan, an analogue of tryptophan, has been studied on the heterocyst spacing pattern and the probability of proheterocyst regression inAnabaena doliolum. 7-azatryptophan suppressed growth and induced heterocyst differentiation in nitrogen-free medium. In ammonium (1 mM), nitrite (2 mM) and nitrate (2 mM) supplemented media, it caused proheterocyst regression with a frequency of 100%, 35% and 10% respectively. The role of azatryptophan in nitrogen metabolism has been discussed in relation to ammonia-uptake study.     

Studies on some enzymes relevant to citric acid accumulation by Aspergillus niger

Enzymatic studies have been performed on a local strain of Aspergillus niger to find a correlation with citric acid accumulation. The activity of aconitase [aconitate hydratase, citrate(isocitrate) hydrolyase, EC 4.2.1.3] and isocitrate dehydrogenase (NADP⁺) [threo-d_s-isocitrate:NADP⁺ oxidoreductase (decarboxylating) EC 1.1.1.42] decreased after 4 days whereas that of citrate synthase [citrate oxaloacetate-lyase (pro-3S-CH₂COO^?→acetylCoA), EC 4.1.3.7] did so after 8 days, when citric acid accumulation in the medium reached a maximum (45.9 mg ml^?1). In vitro studies with mycelial cell-free extracts demonstrated inhibition of citrate synthase activity by sodium azide and potassium ferricyanide on both the 4th and 8th days. Aconitase was inhibited by sodium arsenate, sodium fluoride, iodoacetic acid and potassium ferricyanide only on the 4th day. Isocitrate dehydrogenase (NADP⁺) activity on the 4th and 8th days was inhibited by iodoacetic acid but was stimulated by potassium ferricyanide. The possible existence of isozyme species of these enzymes is discussed.     

NMR spectroscopy in the structural elucidation of oligosaccharides and glycosides.

The potential of one- and two-dimensional NMR techniques for the identification of individual sugar residues, their anomeric configuration, interglycosidic linkages, sequencing and the site of any appended group, in establishing the structures of naturally occurring oligosaccharides and glycosides is presented.     

Evidence for secretion of high molecular weight mucins by canine tracheal epithelial cells in primary culture: Effects of select secretagogues on mucin secretion

Summary The purpose of this investigation was to provide evidence for the secretion of high molecular weight mucins, CTM-A and CTM-B, in primary culture of canine tracheal epithelial (CTE) cells. The cells were isolated from tracheas of mongrel dogs by pronase treatment. Primary cultures of the epithelial cells were established using ICN cellagen inserts in Dulbecco’s modified Eagle’s/F12 medium supplemented with growth factors and could be maintained for up to 23 days. The evidence for the mucin secretion in culture medium and their localization in the cells was established by a) positive immunocytochemical staining using specific antibodies developed against purified native as well as deglycosylated CTM-A and CTM-B; b) incorporation of labeled amino acids, followed by electrophoresis and autoradiography detection of glycoconjugates purified from the culture medium; c) comparison of the amino acid compositions of mucin purified from canine tracheal pouch secretions and that purified from the culture medium; and d) Western blot analyses using specific polyclonal antibodies directed against deglycosylated CTM-A and CTM-B. Immunoaffinity purified secreted labeled glycoconjugates were resistant to hyaluronidase treatment. The effects of cyclic AMP (1 × 10⁻⁵ M), dibutyryl cyclic AMP (1 × 10⁻⁵ M), 8-bromocyclic AMP (1 × 10⁻⁵ M), and prostaglandin E₁ (1 × 10⁻⁶ M) on mucin secretion by CTE cells were also investigated. Secretion of mucins by CTE cells in culture was considerably more enhanced by 8-bromocyclic AMP than that observed for other secretagogues used in this study.     

Hereditary high-adenosine triphosphate syndrome: Study of a new variant

A new variant of hereditary hemolytic anemia in a family due to high adenosine triphosphate (ATP) is reported. The increase in ATP levels varied from 83 to 105% in the family members. Low 2,3-diphosphoglycerate levels and low 2,3-diphosphoglyceromutase activity were observed in three family members, with normal glucose-6-phosphate dehydrogenase and pyruvate kinase activity.     

In vivo phosphorylation of two myelin basic proteins of developing rabbit brain

Examination of 15-day-old rabbit brain myelin proteins by sodium dodecyl sulfate-slab gel electrophoresis revealed the presence of a basic protein with a molecular weight of 21,000 (21K protein) which was not previously reported in this species. This protein exhibited characteristic bluish green color with amido black and gave an amino acid composition strikingly similar to large basic protein (LBP). It formed a line of identity with LBP when diffused against antiserum to chicken brain basic protein. The concentration of LBP (18.9 micrograms/mg of dry myelin) is 6-fold greater than that of the 21K protein(3.31 micrograms/mg of dry myelin) in rabbit brain myelin. After the intracerebral injection of [32P]orthophosphoric acid, both LBP and 21K protein were found to be phosphorylated. [32P]Phosphate in the purified preparations of these proteins was covalently linked by phosphoester bonds to serine and threonine residues. The specific radioactivity of the 21K protein (84,693 cpm/mg of protein) was not significantly higher than LBP (69,797 cpm/mg of protein).     

Accumulation and turnover of the classical Folch-Lees proteolipid proteins in developing and adult rat brain.

The turnover of classical Folch-Lees proteolipid proteins was studied after administration of [2,3-3H]tryptophan to both developing and adult rat brain. The animals were killed from 2h to 250 days after subcutaneous injections of [3H]tryptophan. The measured specific radioactivity in developing brain attained maximum value 24h after the administration of label, whereas the total radioactivity per brain reached a maximum 21 days after injection. The half-life of proteolipid protein from the measured specific radioactivity was 7-20 days, depending on the time-points used for the calculation, whereas calculation from total radioactivity between 28-77 and 91-257 days gave half-lives of 35-40 and 188 days respectively. In contrast, in animals injected at 40 days of age, the half-life from the whole-brain-radioactivity data was 188 days. The problem of the recycling of radioactivity for the synthesis of myelin proteins from either a general or a discrete amino acid pool is discussed.
